In this study, we aimed to evaluate the effects of finasteride on the pharmacokinetics of troglitazone in mice after oral administration. A total of 100 male mice of either sex were used and randomly divided into four groups: Group 1, 10 mg/kg troglitazone (30 mg/kg body weight), Group 2, 5 mg/kg troglitazone (30 mg/kg body weight), and Group 3, 15 mg/kg troglitazone (30 mg/kg body weight). Serial blood samples were drawn at 1, 2, 4, 6 and 12 h after oral administration of troglitazone. The pharmacokinetic parameters of troglitazone were assessed using the bioequivalence of troglitazone in mice after oral administration.
Materials and Methods
After a single oral dose of finasteride (5 mg/kg body weight) was administered, mice were fasted overnight, weighed, and were killed using CO2, and serum samples were collected at the same time points. Troglitazone clearance was measured using a method as previously described by using a noncompartmental method. The mean volume of distribution (Vd) and terminal elimination half-life (T½) were calculated.
After the single oral dose of finasteride (30 mg/kg body weight) administration, mice were fasted for 24 h and weighed.
Conclusions
In the present study, we found that the pharmacokinetic parameters of troglitazone after oral administration of finasteride were significantly different from those in control animals. The pharmacokinetic parameters of troglitazone in mice after finasteride administration were significantly different from those in control mice. The pharmacokinetic parameters of troglitazone after oral administration were higher than those in the control group. The results indicated that finasteride could enhance the pharmacokinetics of troglitazone in mice. These results indicated that finasteride could enhance the pharmacokinetics of troglitazone in mice after oral administration.

Finasteride is a synthetic derivative of the 5α-reductase of the male sex steroid Dihydrotestosterone. It exhibits a strong dihydrotestosterone blocking effect and is used for treating male pattern hair loss in males.
